Fumero has been married to David Fumero, her One Life to Live co-star, since the show’s inception. Enzo, the couple’s first child, was born on March 24, 2016, after the pair married in December 2007. On her last day as a student at NYU, Melissa Fumero joined the cast of One Life to Live as Adriana on January 20, 2004. The post was initially handed to Amanda Cortinas for a few months, but Melissa was then brought in and worked on the project intermittently until 2011. Then, in 2009, she appeared in I Hope They Serve Beer in Hell, Important Things starring Demetri Martin, and Tiny Dancer as Ati, her big-screen debut. She later appeared in Gossip Girl, The Mentalist, Royal Pains, CSI: NY, and Men at Work for the New Year. Finally, in 2013, she joined the cast of Brooklyn Nine-Nine, a comedy television series in which she played Amy Santiago, alongside Andy Samberg in the role of Jake Peralta.

Fumero made her directorial debut on the same series, directing an episode of season 6 of Brooklyn Nine-Nine. She then appeared on Hollywood Game Night, Mack & Moxy, The Admirable, Hell’s Kitchen, The $100,000 Pyramid, One Day at a Time, and the film DriverX after making a guest appearance on Top Chef Duels later that year.
